2 Pharmacodynamics
2.1. Serotonin and NE
Methylphenidate is able to inhibit monoamine oxidase (MAO-A) with IC 50 of 5-30nM, which shows less efficacy at higher concentrations (20nM). This is not due to interfering with the enzyme itself, but instead due to reducing overall efficiency.[44] The inhibition of MAO-A appears to be independent of the catecholamine neurotransmitters; there are no known interactions with monoamines.[44]
When measuring the MAO-A enzyme, a potency of 3.8-5.8µg/mL has been noted, and this inhibition at higher concentrations (up to 160mg/kg), as well the potential involvement in dopamine release was confirmed.[44] This MAO-A inhibition fails to occur with an R-enantiomer (20-hydroxy-5-methylphenidate) a potency of 11.8-20⁺µg/mL (suggesting that the 19.3-40µg/mL concentration is maximum possible inhibition).[44] However, a combination of the two metabolites appears to be equally effective in increasing activity by 15–18%.[44] This is possibly the way methylphenidate acts.[45]
It is thought that MAO-A inhibited, rather than the catecholamines, as inhibiting MAO-A is one of the means increasing dopamine, and is associated with an increase in reward (aldorphins are also known to interfere with MAO).[46]

4.1. Sedation
Methylphenidate has been shown to increase somnolence by increasing both GABA and 5-HT levels, with an increase of GABA being mediated via G-protein activation,[49] and increased 5-HT being mediated by an increase of presynaptic serotonin.[50]
Sedative properties of methylphenidate appear to be mediated via presynaptic G-protein activation.
Methylphenidate is used as a sedative at low doses; the subjective effects are generally not sedative at the doses tested.
In rodents, methylphenidate can be acutely abused by causing seizures.[51] For instance, rats fed methysergide (a reference standard for R-methylphenidate formulations) were found to develop seizures within an hour of treatment. [52]
These seizures are not necessarily toxic, as rats fed the methysergide group survived, while control of rats had to be euthanized on day 3 due to excessive intracranial bleeding. Another rat study, assessing both drugs in the same manner, also showed some effects, at least in rats given the methysergide.[53] It is theorized that this may be due to the amphetamine part of molecule rather than the methylphenidate part, as R-methylphenidate is known to have antihypertensive effects.
